This study evaluated biological responses of cultured murine macrophages (RAW264.7) to PVP-coated silver nanoparticles (nAg-PVP) using assay of WST-8, LDH release, RT-qPCR, and morphological analyses. Cell culture was conducted using traditional two-dimensional culture (2D) and three-dimensional spheroid culture system (3D). The NPs were taken up by receptor-mediated endocytosis, in which particles aggregated in the vesicles. Cell spheroid formation was obviously observed after 4 and s days culture. The cytotoxic effects of cells in 3D were lower than that in 2D.
