To distinguish among pig breeds in raw pork samples, the region of 612 bp corresponding to the domain including second transmembrane of Melanocortin Receptor 1 (MClR) was sequenced in Landrace, Large White, Duroc, Berkshire, Hampshire, Meishan, Vietnamese Moncai and Japanese Wild Boar. Approximately 4 Kbp from exon 16 to 19 of KIT gene was sequenced in European Wild Boar and Yucatan Miniature pig in addition to these eight breeds. Six different sequences with 8 mutated base pairs were obtained from MClR gene. Duroc had a unique sequence as compared to the other breeds, while some group of Landrace, Large White, Berkshire and Hampshire had sequence in common. Sequence obtained from Meishan was identical to that of Vietnamese Moncai, but different from other breeds. Japanese Wild Boars had two different sequences, and both were different from other breeds. In KIT gene, eleven kind of sequences with 62 mutated base pairs were obtained. The mutation from G to A at the position 1, 313 clearly distinguished dominant white color pigs, Landrace and Large White, from other breeds as previously reported. The sequences from Berkshire, Meishan and Japanese Wild Boar were highly similar, as shown by the types of mutation, and 58 of the 62 mutated base pairs were common among the breeds. Landrace, Large White, Hampshire, European Wild Boar and most of Duroc had similar sequences, as indicated also by the types of mutation, and 50 of 62 mutated base pairs were common among these breeds. The sequence observed in several Berkshire from Kagoshima prefecture was similar to that of Yucatan Miniature pigs. This study reveals that the information on polymorphism in the MClR and KIT genes are potent in distinguishing pig breeds that are commercially produced in Japan.
