The interaction between rabbit immunoglobulin G (IgG) and polymethacrylate derivatives, was studied by a microsphere column method. The effects of polymer structure (net charge, hydrophobicity-hydrophilicity, etc.) on adsorption behaviour of IgG was investigated. The results were as follows. (1) The adsorption of IgG to polymethacrylate derivatives (PDEAEMA, PHEMA, PMMA and H. PMMA) was suggested to be mainly supported by hydrophobic interaction and cooperated interactions. (2) Even on the polymeric materials with net charge (PDEAEMA and H. PMMA), the amount of IgG adsorbed ionically was shown to be small. The absorption behaviour of F (ab′) 2 and Fc fragment, digested by pepsin or papain, was investigated in order to examine the orientation of absorbed IgG. The results were as follows. (3) The adsorption of F (ab′) 2 fragment was enhanced by the introduction of cationic sites on polymer surface, conversely as anionic sites was introduced more, that of F (ab′) 2 fragment was lowered but that of Fc was enhanced. The above results suggested that the IgGs can be immobilized on polymer surfaces effectively by designing the characters of polymer surface.
