When tetrodotoxin was injected into the hemocoel of silkworm larva, the small intestine and the rectum exhibited increased contractile movements and the sphinctors at both ends of the colon and the anus were dilated. The hindgut was filled with an uninterrupted string of food which was continuous with midgut contents on one side and with feces on the other. The gut content was transported normally and evacuated by intermittent strong contrations of the rectum. These contractions were abolished by the extirpation of the terminal ganglion or the severance of the posterior lateral nerves. Serotonin, its precursor (5-hydroxytryptophan) and inhibitors of monoamine oxidase (iproniazid phosphate, tranylcypromine) had stimulative effect on the motor activities of the midgut, when they were injected into the hemocoel of larvae paralyzed with tetrodotoxin. Defecation was enhanced by the inhibitors, whose stimulative effect persisted for much longer period than that of serotonin and its precursor. The results indicated that some of the neurons controlling the hindgut maintained the axonal spike propagation in the presence of tetrodotoxin and that serotonin and its derivative acted as neuromodulators for the gut movements.
