Previous study demonstrated that a cell line (ACCS) derived from a human adenoid cystic catcinoma (AdCC) produces a large amount of extracellular matrix (ECM), including basement membrane components, which is resistant to degradation by, and is thus stably accumulated around, the cells. When ACCS cells were plated on either ACCS cell-produced ECM, purified fibronectin (FN), laminin (LAM) or type IV collagen (COLL IV), their migration was strongly enhanced. Motility assays using a modified Boyden chamber showed that FN, LAM and COLL IV stimulate both chemotaxis and haptotaxis of ACCS cells, and their responses are much higher than those of any other tested tumor cell lines. The high motility response seems to be one of the characteristics of ECM-producing tumors such as AdCC, and the findings suggest that AdCC cells use self-produced ECM as a substrate for invasive growth. The special proclivity of AdCC cells to invade nerve and endothelial sheaths may be related to their high chemotactic responses to the basement membrane components. The present study also suggested that both chemotaxis and haptotaxis to ECM, require cellular interaction between integrins and the specific site in the ECM components.
