Proliferation of the biliary epithelium and fibrous tissue associated with hepatolithiasis is the largest problem after repeated treatments. We studied to inhibit the proliferative changes by the gene therapy with direct adenovirus-mediated p 53 gene transfer into the biliary tract in a rat cholangitis model.<BR>To evaluate the effect of the overexpression of p53 gene in a rat cholangitis model, we compared bile duct wall thickness among AxCAhp 53-infected, AxCALacZ-infected, uninfected cholangitis rats. On 7 days after surgery, uninfected and AxCALacZ-infected bile ducts showed a mean thickness of 447.1 ± 53.4μm and 446.5 ± 25.3 du m, and a maximum thickness of 590.0 ± 64.3μm and 650.0±49.3μm, respectively. In contrast, the AxCAhp 53-infected bile ducts showed a mean thickness of 343.2 ± 14.0μm (p<0.05) and a maximum thickness of 408.0±23.9μm (p<0.05).<BR>The Ki-67 Labeling Index were 34.3±4% in AxCALacZ-infected group and 37.4±3.9% in uninfected group, respectively. On the other hand, the AxCAhp 53-infected group was 20.7 ± 5.6% (p<0.05).<BR>p 53 gene transfer with adeno-vector significantly inhibited the proliferative change in the cholangitis model.
