Abstract

The cytotoxicity of PDT with ME2906 and Laser to P388 cells was examined. Cell viability was assayed by the MTT method. When the laser irradiated at 10J/cm<SUP>2</SUP> after incubation for 4 hours with 10μg/ml of ME2906, cell viability was not decreased but with 20 and 30μg/ml of ME2906, cell viabilities were 9 and 1.4% respectively. When the laser irradiated at 10J/cm<SUP>2</SUP> after incubation for 24 hours, cell viabilities were almost same as those of 4 hours incubation. ME2906 contents at 24 hours after drug administration at concentration of 10μg/ml measured 2.8×10<SUP>-9</SUP>μg/cell, and at 4 hours after drug administration at 20μg/ml measured 5.5×10<SUP>-9</SUP>μg/cell. These data showed that the cytotoxicity of PDT to P388 cells was dependent on the ME2906 intracellular accumulated contents but independent on laser doses.
