Abstract

S. parauberisのIa型,Ib/Ic型およびII型に特異的な莢膜合成遺伝子(wzy)の塩基配列からプライマーを設計し,本菌の血清型を識別するマルチプレックスPCR法を開発した。ヒラメ由来S. parauberis 188株について本法を実施したところ,ウサギ抗血清を用いたスライド凝集試験による血清型の判定と一致する成績が得られた。I,II型両抗血清に凝集するために血清型が判定できなかった非特異凝集株もIb/Ic型かII型に判別することができた。ウシ由来S. parauberisを含む他のレンサ球菌や主要魚病細菌は反応しなかった。

A multiplex PCR assay for differentiation of <i>Streptococcus parauberis</i> serotypes was developed. The three primer pairs for subserotypes Ia and Ib/Ic and serotype II were designed from the serotype-specific sequences of the <i>wzy</i> gene in the loci for capsular polysaccharide biosynthesis. All of 188 <i>S. parauberis</i> isolates from Japanese flounder showed positive reaction with the expected size of PCR product for each serotype, which was consistent with the results of agglutination test using rabbit antisera. The nontypeable isolates, which are not differentiated by agglutination test, could be identified as subserotype Ib/Ic or serotype II. Other streptococci including <i>S. parauberis</i> derived from cow and major bacterial fish pathogens showed negative reaction.
