Effect of medium change timing on quality of human iPS cell

Abstract

Purpose: Development of a human iPS cell culture system is required. Culture technology need to be improved to supply iPS cells which is cultured easily and cheaply. In this study, we changed the frequency of medium change and evaluated cell growth and ratio of undifferentiated cells.Method: Cell growth, ratio of undifferentiated cells, pH, glucose and lactic acid were measured when human iPS cells (253G1) were cultured and medium was changed at a frequency of 24 hours and 48 hours.Result: Remaining amounts of glucose were 136 mg/dL (24 hour) and 112mg/dL (48 hour) and increased amounts of lactic acid were 143mg/dL (24 hour) and 140mg/dL (48 hours). The amount of cell growth when the medium was changed at a frequency of 48 hour was 1.3 times of that of 24 hours, which was the standard frequency. In both condition, ratio of undifferentiated cells was the almost same, 95%.Discussion: When the frequency of medium change was low, the cell quality was not decreased and cell growth was improved in short-term hiPS cells culture. This result shows the possibility that hiPS cells are effectively cultured with decreasing the consumption of medium.
