Identification of attached second chromosome of improved strains in the silkworm, Bombyx mori

The linkage analysis and mapping system have been improved and molecular map is now being constructed in the silkworm by using restriction fragment length polymorphism of expressed and sequence tagged cDNA clones. We have tried to identify the silkworm strains, Gensei-kouken, by using these clones mapped on 28 chromosomes. Improved silkworm stains kept at team of molecular development were raised and fourth instar larvae were kept at -40 C. Individual DNA prepared by centrifugation in CsCl gradient were restricted by enzymes and Southern blot hybridizations were carried out. Clones on first chromosome (sex chromosome) were used to identify their sexes. After knowing their sexes, the clones on second chromosome were used as probes to identify the region of attached chromosome. When the clones on the attached chromosome used, female individuals showed same additional band or additional signal intensity but if they are not on attached chromosome, females showed the same pattern to males. By genotyping of the genes on sex, second and attached second chromosomes we tried to identify the strains.
