<p>We have developed a microfluidic device for peptide screening using cancer spheroids. The device washes unbound peptides around spheroids in a droplet using microfluidic approaches such as drainage with capillary pressure, dilution by releasing air between a droplet and washing buffer with pressure barrier, and stirring with circulating flow inside a droplet. The cross-section of the main channel should be enlarged to pass spheroids, though a large cross-sectional channel reduces the capillary pressure and pressure barrier. To satisfy these requirements, the height and width of the main channel is made large and those of the side channel small by the two-layer lithography. The experiment using dyed water demonstrates that the microfluidic device achieves 1/3 dilution.</p>
