The promoter of a H<sub>2</sub>O<sub>2</sub>‐inducible, <i>Arabidopsis</i> glutathione <i>S</i>‐transferase gene contains closely linked OBF‐ and OBP1‐binding sites

<jats:title>Summary</jats:title><jats:p>Glutathione <jats:italic>S</jats:italic>‐transferases (GSTs) are a family of multi‐functional enzymes that play critical roles in the detoxification of xenobiotics and the protection of tissues against oxidative damage. GSTs are important enzymes in plant responses to a number of environmental stresses including herbicides and pathogen attack. <jats:italic>Ocs</jats:italic> elements are a group of related, 20 bp promoter elements which have been exploited by some plant pathogens to express genes in plants. <jats:italic>Ocs</jats:italic> elements have also been found to regulate the expression of a plant <jats:italic>GST</jats:italic> promoter. An <jats:italic>Arabidopsis</jats:italic> GST gene, called <jats:italic>GST6</jats:italic> has been isolated. <jats:italic>GST6</jats:italic> expression is under tissue‐specific control and is induced following treatment with auxin, salicylic acid and H<jats:sub>2</jats:sub>O<jats:sub>2</jats:sub>. The <jats:italic>GST6</jats:italic> promoter contains a binding site for two <jats:italic>Arabidopsis <jats:styled-content>o</jats:styled-content>cs</jats:italic> element <jats:styled-content>b</jats:styled-content>inding <jats:styled-content>f</jats:styled-content>actors (OBF), that has some sequence homology to <jats:italic>ocs</jats:italic> element sequences. Interestingly, OBP1 (<jats:styled-content>O</jats:styled-content>BF <jats:styled-content>b</jats:styled-content>inding <jats:styled-content>p</jats:styled-content>rotein), a DNA‐binding protein that was isolated by screening an <jats:italic>Arabidopsis</jats:italic> cDNA library with a labeled OBF protein as a probe, binds next to the OBF‐binding site on the <jats:italic>GST6</jats:italic> promoter. OBP1 was able to significantly stimulate the binding of OBF proteins to the <jats:italic>GST6</jats:italic> promoter, raising the possibility that interactions between the OBP1 and OBF proteins may be important for <jats:italic>GST6</jats:italic> expression.</jats:p>
