The experiments were carried out to investigate the mechanism of trimoulting of silkworm “Sanminsho” (rt). The perfect supernumerary larval moults were induced by ingesting ecdysterone or applicating juvenile hormone analogue (JHA) during the early stages of the last instar (the 4th instar). A small peak in ecdysteroid titers in the haemolymph of rt larvae was observed at 24hr of the last instar. But at 36hr, it reduced sharply. The changes of juvenile hormone (JH) titers were estimated from the morphological changes induced by injecting ecdysterone, it was supposed that JH titers decreased sharply at 48hr, and disappeared at 96hr of the last instar. The 3rd instar duration of the larvae of rt, which were treated with KK-42 or high temperature and moisture, were prolonged, and the perfect supernumerary larval moult could not induced by application of JHA during the early stages of the 4th instar of these treated larvae. There was not such a small peak in ecdysteroid titers in the haemolymph of the treated larvae as the control larvae at 24hr of the last instar. From these results, it was supposed that the endocrine system directing pupal metamorphosis has not been determined during the early stages of the last instar of rt, and the decrease of ecdysteroid titers at 36hr of the last instar might play an important role in trimoulting of rt.
