Metabolism of amino acid in the canine warm ischemic liver and effect of Prostaglandin E1(PGE11) on the metabolism were studied.<BR>Mongrel dogs were divided into three groups; control group and groups of liver ischemia with or without PGE1 pretreatment. Liver ischemia for 60 min was induced by Pringle's method. PGE11 was intravenously administered before Pringle's procedure for the group of ischemia with PGE1. Total free plasma amino acid(TFPAA), aromatic amino acid(AAA), branched chain amino acid(BCAA), BCAA/AAA and serum lipid peroxide concentrations in portal, hepatic and peripheral venous blood were examined at the points before clamping, 5, 60 and 120 min after declamping. In addition, liver metabolic rate of above mentions substances at the corresponding time was examined.<BR>The metabolic rate which was calculated by formula as follow: (portal venous concentrationhepatic venous concentration)/portal venous concentration.<BR>In the group of ischemia without PGE1 pretreatment, the metabolic rate of TFPAA, BCAA/AAA in hepatic venous blood were decreased 5 min after declamping, and TFPAA, AAA, BCAA and lipid peroxide concentrations in portal, hepatic and peripheral venous blood were increased after the declamping. In the group of ischemia with PGE1 pretreatment, however, the TFPAA metabolic diturbance in the ishcemic liver was improved, and increased TFPAA, AAA, BCAA and lipid peroxide concentrations by the warm ischemia were normalized.<BR>This study suggested that PGE1 protects warm ischemic liver damage resulting in improvement of metabolic disturbance of amino acids and depression of lipid peroxide production.
