This discusses new and established strategies for delivering peptides and antibodies through the blood-brain barrier. It reviews concepts on blood-brain barrier transport biology, assesses the utility and limitations of traditional brain drug delivery methods and describes new drug delivery techniques. The opening section of the book presents a review of neuropeptide biology and the potential of clinical applications of neuropeptides. Next comes information on the principles of blood-brain barrier transport physiology and cell biology. Attention is given to the problems involved in developing blood-brain barrier transport vectors that are capable of shuttling neuropeptides or monoclonal antibodies across the blood-brain barrier and into specific areas of the brain. Subsequent chapters review existing modalities for drug delivery to the brain including neurosurgical-based and pharmacologic-based strategies. The second half of the book focuses on strategies for blood-brain barrier delivery of peptides, antibodies and other neuropharmacuticals.
These include methods for : receptor-mediated and absorptive mediated transcytosis of peptides and antibodies through the blood-brain barrier; delivery of chimeric peptide neuropharmaceuticals through the blood-brain barrier via chemical linkers, avidin-based linkers and peptide linkers formed in gene fusions; and molecular biological strategies for the development of brain-specific transport vectors. Some 900 citations are lised, many from papers published in 1990.
