This volume is a comprehensive review of the biology of tumour necrosis factor (TNF) lymphotoxin, two cytokines that cause a vast array of changes in cell behaviour in virtually every tissue and play a major role in a wide variety of disease processes, from endotoxic shock to bone resorption. More than 60 leading investigators in diverse basic science and clinical disciplines synthesize discoveries on the biosynthesis of these cytokines, their molecular structure, their signal transduction mechanisms in cells, their interactions with other cytokines, their effects on various tissues and organ systems, their tumor necrotizing activity, and their physiological and pathophysiological functions. The findings presented indicate that, while TNF can cause endotoxic shock, cachexia, inflammation, and bone resorption, it can also protect against the lethal outcome of various infections and attenuate the development of certain autoimmune diseases. The contributors also explore the potential medical uses of TNF and TNF inhibitors.
