Since the publication of the first edition of "Cytochrome P-450" in 1978, knowledge of this unique hemoprotein has grown considerably. A large number of different forms of cytochrome P-450 have been isolated from animal tissues, plants and microorganisms and much information on their molecular properties and physiological functions has accumulated rapidly. It is now firmly established that cytochrome P-450 is the collective name for a large group of hemoproteins having the same ligand structure around the heme and that they play highly important roles in the metabolism of various endogenous substrates in animals, plants and eukariotic microorganisms. In animal tissues, various forms of cytochrome P-450 play central roles in the biosynthesis of cholesterol, steroid hormones and bile acids, the metabolism of fatty acids and eicosanoids as well as in the metabolism of xenobiotic compounds including drugs. Cytochrome P-450 is thus deeply involved in various metabolic pathways essential for the maintenance of the life of higher organisms. The aim of this volume, the second edition of "Cytochrome P-450" is to provide the reader with an up-to-date survey of the present knowledge about this unique group of physiologically important hemoproteins. In order to cope with the new developments in the research on cytochrome P-450 since the publication of the first edition, the organization of the book has been changed, and all the chapters have been newly written by expert contributors. A short history of the research on cytochrome P-450 is given in chapter one as an introduction to the following chapters and the basic molecular properties of cytochrome P-450 and the mechanisms of cytochrome P-450-catalyzed reactions are described in chapter two. Chapter three emphasizes the physiological functions of various forms of cytochrome P-450 in animal organs. Chapter four is a concise account of the occurence and functions of cytochrome P-450 in eukaryotic and prokaryotic microorganisms. Chapter five describes recent developments of the research on the structures of cytochrome P-450 genes and their expression on mammalian organs. The last chapter discusses the evolution and differentiation of cytochrome P-450 genes. A commentary on the nomenclature of various forms of cytochrome P-450 with a complete list of the hemoproteins are given in the appendix.
