This work is based on lectures given at the Ettore Majorana Centre's 25th Course on "Resistance to Anti-tumour Agents in the Laboratory and Clinic: Problems and Implications from 13 to 18 April 1988". In the last few decades extensive research worldwide has led to the discovery of a number of compounds with properties that inhibit the proliferation of neoplastic cells. These compounds make up a large array of therapeutic agents now available to the physician treating cancer patients. Experimental evidence and controlled clinical trials, however, have shown that the failure to reach expected goals during treatment is due principally to the emergence of drug resistance cancer cell clones. The neoplastic cell has multifarious pathways options, enabling its survival and multiplication in the presence of inhibitors. The therapist's task is to design regimens using single or combined treatments in order to prevent or circumvent the obstacle of drug resistance. This book is intended to bring clinical oncologists up to date with the problem and to stimulate further work in the field.
