This book is about the determination of the molecular structures of protein toxins and the impact that this work has had on the understanding of toxin action. The key technique used in these studies has been X-ray crystallography. Protein crystallography is the study of the three-dimensional shapes of proteins at near atomic resolution. Toxins are excellent model systems for understanding the insertion and translocation of proteins across cellular membranes. Furthermore, studies on pore-forming toxins are providing insight into membrane protein biogenesis and the molecular architecture of membrane ion channels. Of particular interest to toxicologists is the possibility of rationally designing drugs using a knowledge of the three-dimensional structure of key protein toxins so as to inhibit bacterial virulence.
