Until the '90s, research in biomechanics and biorheology mainly focused on tissues and cell priorities, so as to identify the bioclinical problems linked to the rheology properties of cells and tissues or to develop substitutes. Advances in molecular biology and new knowledge in cellular biology over the last ten years have given access to a more physiological approach to the effects of stress on cells and tissues. All cells and tissues in the body are constantly exposed to physical forces and these forces can influence the biological behaviour of cells and hence affect gene expression, phenotype, paracrine or autocrine factor secretion and metabolism. These mechanically-induced cellular alterations may constitute major factors affecting the physiological and pathological condition of the organism. This new approach could be called mechanobiology because it requires, in parallel with theoretical concept development at the cellular level, the use of molecular biology methods to identify the various steps or stages whereby changes are brought about by the mechanical forces permanently applied to cells and tissue.
This book focuses on cartilage and chondrocytes, which are the cells particularly subject to wide range pressure stress.

The main aim of this book is to focus on research in the mechanobiology of cartilage and chondrocyte, and to promote the creation of new studies and collaborations in the osteo-articular field. It is important to focus on the different mechanisms which arise during cartilage remodelling and the possibilities of clinical applications. Three main themes were developed during the two days' work in the Paris symposium: the mechanical propoerties of cartilage and their modelling; the importance of the effects of mechanical stress on the physiology of the chondrocyte; and clinical applications and cartilage engineering. For future work there is a need to reinforce clinical and pharmacological studies on the importance of mechanical stresses on the properties of cartilage.

Mechanobiology is now a vigorous branch of biomechanics and biorheology which is mainly concerned with the study of the influence of mechanical forces on cells and tissues and their clinical or therapeutical applications. As we are now at the age of proteomics and genomics and of cell micromechanical approaches, using methods like laser tweezers or confocal microscopy, mechanobiology brings new challenges. With these new researches, mechanobiology is the promise of new diagnostic and therapeutic approaches. T he most recent work shows that the incidence of mechanical forces is specific to the system under scrutiny and that stresses are implicated in tissue physiology (for example by the production of the extracellular matrix), secretions (i.e. production of NO and prostaglandins by endothelial cells), or for the induction of specific functions via intercellular communication; hence the interest from pharmacology in studies on new molecules.
Moreover, these new findings have led to the development of tissue engineering, which is the concept of substitute tissue developed in vitro, from bioresorbable or non-bioresorbable scaffolds and from cells harvested in a physiologic mechanical environment such as from cartilage, bone and vessels. At the same time, the problem of cell grafting in tissue repair and especially the use of stem cells have led to new therapeutic fields.

This book covers the proceedings of the Fifth Symposium on Mechanobiology of Cartilage and Chondrocyte. Mechanobiology can be now considered as a vigorous branch of biomechanics, biorheology and physiology mainly concerned with the study of the influence of mechanical forces on cells and tissues and their clinical or therapeutical applications. As we are now in the age of proteomics, genomics and cell micro mechanical approaches, suing methods like laser tweezers or confocal microscopy, mechanobiology brings new challenges. With such new research, mechanobiology promises new diagnostic and therapeutic approaches. In other respect there has been increasing interest over recent years in the fundamental role played by local mechanical parameters in chondrocyte regulations and cartilage dysfunctions as a first step in the development of osteoarthritis. These proceedings are sub-divided into four parts: Theoretical approaches and mechanobiology of chondrocyte; Cartilage and chondrocyte studies; Osteoarthritis: inflammation degradation and clinical approaches; and, Cartilage engineering.
